I think this best sums it up for me.

When Alex and Justin Smith won the Eagles game.

I always thought they were the best team in the division, based on talent. But when Alex did that on the road, in an early game, with that deficit... I knew this team had a chance to be special. Superbowl special.
I agree with all of the above; the Eagles game signified A LOT of things. -We were coming off of a rough/uneventful road win against a growing Bengals squad, walking into Philly with all odds against us, never having won a game there in practically a decade (nearly never having won a game period against the eagles in a decade, itself)...

And then we do it... we stage one of the biggest single-season comebacks since probably 1980 against the Saints... and the moment Justin Smith knocked that ball out of Jeremy Maclin's arm... I was in tears... I knew things were gonna change... I knew things were different this time...

Philly.
A lot of people say that they beat a bad Eagles team in a comeback. That may be so but every team's turning point is different. It may not be necessarily against a good team but that can't dismissed the comeback itself. I remember the '81 team had one of the biggest come back against the Saints AKA the Aints. Or the '94 team after being annihilated by the Eagles at home. Being down 14-0 against the Lions in their dome, with Steve Young crawling out up from a sack. The team came back to win that game and never look back.

For the fans including myself it would be the Giants game. But for the players it was earlier in the Eagles game.
i made this predicition after week 1 vs the seachickens. i have the post to prove it.

Yup Giants game confirmed my beliefs

That's the one. When the 49ers beat the Eagles, I thought that luck might be on their side, but the game in Detroit confirmed that they were for real, and Smith could definitely come from behind to win consistently.
Playoff contender - When we beat the eagles. Any of the Nolan and Singletary teams would have folded.

SB contender - When we beat the Giants. They were hot coming off a win against the patriots, and we won with the
nation watching.
I thought they were for real after the Lions game, but a SB contender? I had to wait until the Giants game to know for sure they are a true contender.

And that was also Detroits coming out party... they win and they are solidified as the next great team on the rise. We lose, we had a good excuse, good lions team, crazy loud stadium, east coast time zone.. etc. But they went in and beat the lions at what was supposedly the strongest aspect of their team, the D-line. Keeping CJ on lock from the end zone was also impressive. This team just went about their business and won a game that would have been a blow out in previous years.
I swear the day Jim Harbaugh signed, I straight up said "CHAMPIONSHIP!"

no one payed attention to me.
